McMaster University researchers have made a groundbreaking discovery in the fight against brain cancer. The team, led by Sheila Singh and Jakob Magolan, has developed a novel therapeutic approach that could revolutionize the prevention of cancer metastasis to the brain.
The study, published in the Proceedings of the National Academy of Sciences, introduces a new class of drug candidates designed to target a specific enzyme, IMPDH2, which plays a crucial role in the development of brain metastases. Unlike previous attempts to target IMPDH, this approach focuses on IMPDH2, which is not abundant in healthy tissue, allowing for more selective and effective cancer cell elimination.
Singh emphasizes the grim outlook associated with metastatic brain cancer, which is the most common type of brain tumor in adults. Current treatments are largely palliative, making the development of preventative interventions a priority. By identifying high-risk patients and intercepting metastatic cells before they can form brain tumors, the team aims to transform this fatal disease into a preventable one.
The drug candidates are being developed by Block Biosciences, a McMaster spin-out company. The collaborative effort between medicinal chemists at McMaster and Block has led to the design and synthesis of several hundred IMPDH2-targeting drug candidates. The team is now selecting the most promising compounds to advance in clinical development.
Magolan expresses optimism about the clinical potential of the top drug candidates. These compounds possess essential properties, such as staying in the body long enough to be effective, crossing the blood-brain barrier, and synergizing with existing cancer medicines. However, further refinement is needed before human trials can commence.
